Impact of 2-hydroxyglutarate-induced astrocytes reactivity in low-grade glioma progression and recurrence

Mutations in the metabolic enzymes are commonly observed in brain cancers. Although the precise molecular underpinning for this remains elusive, it has been shown that brain cancer-associated mutations in metabolic pathways confer altered metabolic function by increasing production of the oncogenic metabolite R-2-Hydroxyglutarate (R-2HG). This metabolite was shown to promote tumor progression to a more aggressive form of brain cancer and thus, was termed « oncometabolite ». Here we propose to determine how metabolic defects associated with the production of this oncometabolite leads to the establishment of a proliferative environment that promotes tumour progression and facilitate its recurrence, a totally novel concept in molecular oncology.
